ABSTRACT

BACKGROUND: In spring 2021 increasing numbers of cats presenting with severe pancytopenia were noted in United Kingdom (UK). OBJECTIVE: To describe process and outcome of the investigation performed into the outbreak of pancytopenia in cats. ANIMALS: Five hundred and eighty client owned cats that presented with severe bi- or pancytopenia of unknown cause. METHODS: Real-time data collection was performed by an online registration forum available to all veterinary surgeons in UK. Data collected included demographics, clinicopathological findings, diagnostic testing, dietary and drug history, outcome and COVID household status. Mycotoxicological feed analysis was performed on feed samples of 3 diets frequently mentioned in the database and 3 control diets. RESULTS: Five hundred and eighty cats presented to 378 veterinary practices were included for analysis. Case fatality rate was 63.3%. Dietary history was available for 544 (93.8%) cats, of which 500 (86%) were fed 1 of 3 diets (which were recalled midinvestigation). 54 (9.3%) cats were not fed a recalled product, with diet information unknown in 26 (4.5%) cats. Analysis of feed samples revealed concentrations of hematotoxic trichothecene T-2/HT-2 mycotoxins greater than recommended by the European Commission in 5/7 recalled diet samples but in none of control diet samples. The trichothecene mycotoxin diacetoxyscirpenol (DAS) was detectable in all recalled diet samples but not in any of control samples. CONCLUSION AND CLINICAL IMPORTANCE: Contaminated-feed induced trichothecene mycotoxicosis should be considered as a differential diagnosis for pancytopenia in cats.

ABSTRACT

Broiler population is one of the most important segments of livestock due to its significant contribution in white meat production. Infectious disease outbreaks adversely influence the production potential and consequently cause economic losses. Epidemiological data regarding magnitude of these disease outbreaks is of fundamental importance for planning of a comprehensive control strategy. With retrospective design, this study was conducted from January 2013 through December 2017 in order to assess the disease burden on broilers reared in different open type poultry houses. Out of total 658 commercial farms with capacity of 4221800 broilers, across Chakwal, a representative sample of 70 farms with capacity of 448000 broilers was randomly selected for collection and analysis of disease data. Five years' data of these randomly selected farms revealed highest (44.64%) crude morbidity during monsoon season followed by 23.92%, 22.12% and 17.49% for winter, spring and post-monsoon seasons respectively. The highest (14.90%) prevalence was recorded for new castle disease followed by infectious bursal disease (11.79%), pullorum disease (11.17%), colibacillosis (8.71%), infectious bronchitis (7.87%), inclusion body hepatitis (7.79%), chronic respiratory disease (7.67%), necrotic enteritis (6.48%), coccidiosis (6.09%), mycotoxicosis (5.43%), fowl cholera (4.74%), infectious coryza (4.41%), fowl typhoid (4.22%), omphalitis (3.71%) and hydropericardium syndrome (0.05%). Maximum share in crude morbidity was contributed by bacterial diseases with highest proportional morbidity of 48.68% followed by viral (40.32%), parasitic (5.80%) and fungal (5.20%) diseases. This epidemiological data represents true picture of study population and is a valuable tool for planning of prevention strategy and research priorities.

ABSTRACT

A two-year (2018/19 and 2019/20) field experiment was carried out to evaluate the efficacy of recently developed fungicide combinations (with different modes of action) towards fungal diseases on seven bread and eight durum wheat varieties. The trial was performed at the FIELDLAB experimental station of the University of Perugia (Italy). The diseases were assessed under natural pressure except for Fusarium head blight (FHB) for which artificial inoculation with a Fusarium culmorum deoxynivalenol (DON)-producing strain was performed at the full flowering stage (BBCH 65). Fungicides were sprayed at the fully extended flag leaf (BBCH 39) and full flowering (BBCH 65) stages. The incidence of different fungal diseases was visually evaluated and other parameters [grains production (t/ha), protein content (%), test weight (kg/hL), and DON accumulation in grain (μg/kg)] were also determined. In the two years, characterized by different climatic conditions, the fungicide treatments showed efficacy in controlling the observed diseases (Septoria tritici blotch and FHB) as well as in reducing DON contamination. No significant differences were found between treatments. The results highlight that, in the present scenario of commercially available durum and bread wheat varieties, the timely application of the most common fungicides plays a crucial role for FHB and DON management in the presence of climatic conditions that are favorable to the disease. The impact of these results in an integrated disease management perspective is discussed. ABSTRACT

Mycobiota is not only associated with healthy homeostasis in the human gut but also helps to adapt to the environment. Food habits, alcohol consumption, intake of probiotics, and contaminated food with a mycotoxin, often lead to the alteration in the mycobiota composition. Impaired immunity of the host may affect fungal symbiosis leading to mycosis. The human gut adapts to the commensalism fungi belonging to the phylum Ascomycota and Basidiomycota. Diet habits such as plant-or animal-based, phytoestrogens enriched plant products, fat-rich diets also influence the colonization of certain fungal species in the mammalian gut. Food habits or mycotoxin-contaminated food or fungal peptides have an impact on bacterial-fungal interaction and human health. The mycobiota population such as Fusarium, Humicola, Aspergillus, and Candida are altered due to alcohol intake in alcoholic liver disease. The role of associated gut mycobiota due to irregular bowel habits or lifestyle change has been observed in inflammatory bowel disease. In this review, it has been observed that Saccharomyces, Aspergillus, Fusarium, Cladosporium, Candida, and Malassezia were the common genus in the human mycobiota. Therefore, this study focused on how diet habits and alcohol intake, among others., influence mycobiota composition that may affect the human immune system or overall health.

ABSTRACT

Coix seed is an important food and traditional Chinese medicine in China and other Asian countries. Notably, coix seed is currently being used as a traditional medicine for the treatment of COVID-19 in China. However, coix seeds are generally contaminated by mycotoxins, and this risk cannot be ignored. In this paper, we developed a method that involves direct extraction and UHPLC-HRMS analysis for the simultaneous detection of 24 mycotoxins in coix seeds. UHPLC-HRMS instrument and data acquisition parameters, and the sample pretreatment were optimised. One-step extraction showed several advantages compared to the three commercial solid-phase extraction clean-up methods, including ease of use, reduced time of sample preparation, low cost, good recovery, and acceptable matrix effect. The method validation results indicate that all mycotoxins have good linearity and sensitivity. Recoveries were between 74.2-101.1%, and RSD ranged from 0.1-5.8%. The LOQs for 24 mycotoxins were in the range of 0.5-100 µg/kg. To survey the contamination levels of these mycotoxins in commercial coix seeds, more than 70 samples were collected from Chinese markets and were analysed using the newly developed method. Zearalenone (positive ratio: 98.7%, range:1.1-1562 µg/kg), deoxynivalenol (positive ratio: 87%, range: 8.4-382.5 µg/kg), nivalenol (positive ratio: 85.7%, range: 26.8-828.2 µg/kg), fumonisin B1 (positive ratio: 84.4%, range:2.5-314.5 µg/kg), fumonisin B2 (positive ratio: 75.3%, range:1.6-72.8 µg/kg), fumonisin B3 (positive ratio: 48%, range:1.0-203.6 µg/kg), aflatoxin B1 (positive ratio: 29.9%, range: 0.39-14.7 µg/kg), sterigmatocystin (positive ratio: 29.9%, range: 1.4-51.6 µg/kg), and tenuazonic acid (positive ratio: 19.5%, range 36.1-105.7 µg/kg) were the most frequent mycotoxin contaminants. These results highlight the importance of routine monitoring and control of mycotoxins in coix seeds.

ABSTRACT

A feeding trial (35 days) was carried out to investigate the effect of Saccharomyces cerevisiae cell wall as a mycotoxin biodegradation agent on the performance, feed efficiency, carcass traits, and immunity response against diseases in broilers fed aflatoxin B1 contaminated diets. For this purpose, 200 one day old broilers were randomly allotted into four groups, each with five replicates (10 birds per replicate). Four starter and finisher experimental rations were formulated by using (A) 0, (B) 1.25, (C) 2.5, and (D) 3.75 g kg-1 of Saccharomyces cerevisiae. Experimental diets were contaminated with aflatoxin B1 (100 ppb kg-1 diet). The experimental chicks were kept under standard managerial conditions, and the vaccination program was followed against infectious bursal disease (IBD), infectious bronchitis (IB), and Newcastle disease (ND) diseases. At the end of the feeding trial, carcass, organ weight, and blood samples were collected randomly to determine the carcass traits and antibody titer against ND and IBD viruses. Throughout the experiment, the addition of 3.75 g kg-1 of the Saccharomyces cerevisiae cell wall (Group-D) in feed resulted in the highest weight gain, final weight, feed intake, and the lowest FCR values followed by C group compared with the other groups. All carcass traits were significantly (p > 0.05) improved by increasing the inclusion levels of Saccharomyces cerevisiae in broiler diets. It could be concluded that the broiler diet supplemented with 2.5 or 3.75 g kg-1 of Saccharomyces cerevisiae as a biodegrading agent resulted in improved growth performance, immunity activity and carcass traits, and supplementation with Saccharomyces cerevisiae at these levels can be used effectively in broiler diets without negatively affecting bird health status.
